ABSTRACT

Objective:To investigate the efficacy of anti-CD 20 monoclonal antibody in combination with Gemcitabine-based chemotherapy regimen in children with recurrent diffuse large B-cell lymphoma (DLBCL). Methods:The clinical data of 62 children with DLBCL admitted to the Department of Pediatrics, Yantai Mountain Hospital of Yantai City and the Department of Pediatrics, Muping District Traditional Chinese Medicine Hospital in Yantai City from January 2010 to January 2018 were analyzed retrospectively.Different treatment options were selected according to the children′s stage and the presence of risk factors such as huge tumors.Among them, 32 cases in the control group were treated with the Gemcitabine-based treatment plan (Gemcitabine+ Cisplatin+ Dexamethasone treatment). Thirty patients in the study group were treated with anti-CD 20 monoclonal antibody on the basis of the control group for a total of 4 cycles (21 d per cycle). After 4 cycles of treatment, the clinical efficacy, the positive expression of forkhead box protein P1 (FOXP1) and B-cell lymphoma factor-6 (Bcl-6) before and after treatment, the occurrence of adverse reactions and survival[3-year progression-free survival (PFS), 3-year overall survival (OS)] were evaluated.The measurement data that meets the normal distribution is expressed that the t test is used, and the counting data is represented by (%), and the χ2 test is used.Level data is compared with ranking and inspection. Results:All patients were followed up to March 2021.The total response rate (RR) and disease control rate (DCR) of the study group were 93.33% (28/30 cases) and 96.67% (29/30 cases), respectively.The RR and DCR of the control group were 68.75% (22/32 cases) and 81.25% (26/32 cases), respectively.The RR of the study group was higher than that of the control group ( χ2=5.995, P<0.05), and there was no statistical difference in DCR between the two groups ( χ2=3.674, P>0.05). After treatment, the positive expression rate of FOXP1 in the study group and the control group was lower than before treatment[23.33% (7/30 cases) vs. 76.67% (23/30 cases); 50.00% (16/32 cases) vs. 75.00% (24/32 cases), χ2=17.067, 4.267, all P<0.05], and the positive expression rate of the study group was lower than that of the control group ( χ2=4.179, P<0.05). After treatment, the positive expression rate of Bcl-6 in the study group and the control group was higher than before treatment[86.67% (26/30 cases) vs. 26.67% (8/30 cases); 62.50% (20/32 cases) vs. 31.25% (10/32 cases), χ2=21.991, 6.275, all P<0.05], and the positive expression rate of the study group was higher than that of the control group ( χ2=4.723, P<0.05). There was no significant difference between the study group and the control group in the level of gastrointestinal reactions, elevated transa-minase, and decreased white blood cell ( Z=-1.074, -1.078, -0.834, all P>0.05). There was a difference between the 3-year PFS survival curve and the 3-year OS survival curve between the two groups ( χ2=3.997, 4.723, all P<0.05). Conclusions:Anti-CD 20 monoclonal antibody combined with Gemcitabine-based chemotherapy is effective for children with DLBCL, and will not significantly increase the adverse reactions in children.

ABSTRACT

This study was aimed to investigate the effect of Forkhead Box G1 (FOXG1) on the epithelial-mesenchymal transition (EMT) of colorectal cancer (CRC) cells and the underlying mechanism. For this purpose, FOXG1 lentiviral interference (shRNA) plasmid and expression plasmid were constructed. Western blotting was used to analyze the expression of FOXG1 protein in five CRC cells, namely RKO, SW480, SW620, LoVo and DLD-1. The shRNA fragment of FOXG1 (shFOXG1) was designed and synthesized. Recombinant plasmids were obtained with the aid of DNA recombination technique. Double digestion and sequencing were used to identify the recombinant plasmids, and then lentivirus packaging, purification and stable transfection were carried out. Additionally, stable CRC cell lines were screened out. The changes of FOXG1 knockdown and overexpression efficiency, E-cadherin, Vimentin, Fibronectin, Snail, Twist mRNA and protein were investigated respectively by Western blotting and qRT-PCR analysis. Furthermore, the changes of cell morphology after knockdown and cell migration ability were evaluated respectively with optical microscopy, scratch test and Transwell assay. FOXG1 had the highest protein expression in RKO and the lowest in DLD-1 among the five CRC cells. Compared with those of the control group, the cell morphology in FOXG1 knockdown RKO group was changed from spindle into round or polygonal shape, cell polarization was enhanced and tight junction assembly was acclerated while cell migration distance was noticeably decreased. Moreover, the number of cells invaded and migrated through chambers was significantly reduced. Among these key factors of EMT, the expression of E-cadherin was increased while the expressions of Vimentin, Fibronectin, Snail and Twist were decreased. The opposite was the case in the overexpressed FOXG1 group. The overexpression of FOXG1 in CRC promoted the invasion and metastasis of CRC cells and played a crucial role in regulating the EMT. Thus, FOXG1 might be a novel therapeutic target in CRC treatment.

ABSTRACT

Objective To investigate the application of damage control surgical treatment in the patients with severe abdominal infection and its nursing experience. Methods Toally 10 patients with severe abdominal infection were treated with damage control measures, including damage control surgery, improvement of pathological and physiological status of patients and surgical re-repairing, and the corresponding nursing measures including rapid completion of preoperative preparation, prevention of complications, nursing care to abdominal double cannula drainage, enteral nutrition and disease observation and mobilization the subjective initiative of patients. Results The average hosptitalization time was 21 days (14~29 days). There were 2 cases of anastomotic leakage and 1 case of pulmonary infection who all were cured after 14~29 d. Conclusion Damage control surgery is an effective treatment strategy for patients with severe abdominal infection and the corresponding nursing plays a positive role in improving the treatment effect.

Objective To analyze the ecological environmental differences of hibernation hole and temporary hole of Himalayan marmot based on global positioning system (GPS),remote sensing (RS) and geographical information system (GIS) in Yushu City,Qinghai Province.Methods Hibernation holes and temporary holes were located based on GPS.Ecological environment factors related with Himalayan marmot,such as land surface temperature (LST),digital elevation model (DEM),normalized different vegetation index (NDVI),were extracted based on RS and GIS.The ecological environment differences between hibernation hole and temporary hole were explored based on epidemiological statistical method.Results Monthly LST (degree Kelvin,K) of hibernation hole was slightly higher than that of temporary hole in the same period.Yearly day LST of hibernation hole was 1.1 K higher than that of temporary hole in the same period.Yearly night LST of hibernation hole was 0.3 K higher than that of temporary hole in the same period.Monthly NDVI of temporary hole was slightly higher than that of hibernation hole in the same period.Yearly NDVI of temporary hole was 0.001 higher than that of hibernation hole in the same period.DEM of temporary hole was about 40 m higher than that of hibernation hole.The top two ratios of soil type factors were both alpine meadow soil and dark felty soil for hibernation hole and temporary hole,which were 62.1% and 29.3%,64.4% and 25.6%.The top two ratios of land cover factors were both rainfed cropland and mosaic cropland for hibernation hole and temporary hole,which were 43.3% and 25.8%,40.0% and 26.2%.Conclusions Soil type factors and land cover factors for hibernation hole and temporary hole are similar.Lower DEM and higher LST are chose for hibernation hole,and better NDVI is chose for temporary hole,which is good for food intake.

ABSTRACT

Objective To compare the levels of serum phosphorus,bone alkaline phosphatase and 25-hydroxyvitamin D3 in preterm infants with very low birth weight, and provide evidence for early screening, prevention of metabolic bone disease in preterm infants.Methods A total of 110 newborns who met the inclusion criteria were selected in pediatric ward in our hospital.The case group included 60 preterm infants with very low birth weight and control group included 50 full term infants.Fasting blood were taken from the subjects at week 1, 4, and 12 respectively, and ELISA was conducted to quantitatively detect the serum levels of phosphorus, bone alkaline phosphatase and 25-hydroxyvitamin D.Results The increase of serum 25-hydroxyvitamin D3 in case group was smaller than that in control group;the levels of serum phosphorus had no significant difference between two groups.In case group, the level of bone alkaline phosphatase increased at week 1,4, and 12, and the 25-hydroxyvitamin D3 level had significant difference at week 12 (P<0.05).The abnormal rate of 25-hydroxyvitamin D3 level was 26.7% for case group and 0% for control group.Conclusion There were significant differences between case group and control group in levels of bone alkaline phosphatase and 25-hydroxyvitamin D3, suggesting that detecting the serum levels of bone alkaline phosphatase and 25-hydroxyvitamin D3 would facilitate the early diagnosis of metabolic bone disease in preterm infants.

ABSTRACT

Obgective To investigate the impact of different ways and doses of recombinant human interferon o1b(rhIFN-α1b) on antiviral effect on respiratory syncytial virus(RSV)-effected mice and signal pathway of JAK/ STAT.Methods Forty-eight mice were divided randomly into 6 groups,negative control group,RSV-infection model group,12.5,25.0,50.0 μg rhIFN-α1 b atomization inhalation of intervention group,and 12.5 μg rhIFN-α 1 b injection of intervention group.After continuous drug therapy for 5 days,the left lung tissues of mice were aseptically dissected in the sixth day.Then it were observed lung tissue pathology changes by optical microscope,and expression of STAT1 and STAT2 protein with laser scanning confocal microscopy.Results It was observed that RSV-infection model mice's lungs had significant inflammatory injury under light microscope.After treatments of rhIFN-α 1b,it showed that the mice lung tissue had recovery of inflammation on different degrees.The group of inhaled rhIFN-o1b 50.0 μg was damaged on lightest degree.There were statistically significant differences between each group (all P < 0.05).The expression of STAT1 and STAT2 protein in RSV-infection model mice's lungs decreased remarkably under fluorescene microscopy.Treatments of rhIFN-α 1b increased the expression of STAT1 and STAT2 inhibited by RSV-infection.The expression of STAT1 and STAT2 protein of the inhaled rhIFN-α1b 50.0 μg group increased significantly.The differences between groups were statistically significant(all P < 0.05).Conclusions rhIFN-α 1 b played an important role in anti-RSV effects,simultaneously,it could improve the activity of JAK/STAT signal pathway inhibited by RSV-infection.The treatment effect of rhIFN-α 1b deliveried by atomization inhalation was better than that of intraperitoneal injection.And curative effect is proportional to the atomization inhalation dose within a certain range.

ABSTRACT

Background and purpose:Single drug of docetaxel and pemetrexed as second line treatment is standard treatment of advanced non-small cell lung cancer (NSCLC). Whether combined with platinum can increase the response and survival is still not elucidated. This study was designed to investigate the treatment response, overall survival (OS) and the safety of combined with oxaliplatin or cisplatin regimens as second line in treating NSCLC patients. Methods:Advanced NSCLC inpatients, failure of cisplatin or carboplatin in initial treatment, were divided into three groups at random in 3∶2∶1 rate. Control group:who received docetaxel, 75 mg/m2 (for all patients), d1 or pemetrexed 500 mg/m2 (for non-squamous carcinoma);Cisplatin group:who received cisplatin 25 mg/m2, d1-3 and docetaxel/pemetrexed; Oxaliplatin group: who received oxaliplatin 130 mg/m2 d1 and docetaxel/pemetrexed. Every 3 weeks were repeated as one cycle. The side effect was assessed every cycle and treatment efifcacy was investigated every two cycles. Follow-up examination was taken every 3 months after treatment. Results:There were no differences in treatment response, progress free survival (PFS), OS and toxicity among the three groups (P>0.05). Old patients (≥60 years) had a better PFS than that of patients less than 60 years (HR=0.56, 95%CI:0.35-0.90, P=0.015). Patients with performance score 0-1 had a better PFS and OS (HR=1.52, 95%CI:1.01-2.30, P=0.048;HR=1.90, 95%CI:1.17-3.09, P=0.009). Treatment response had relation to PFS and OS (HR=2.93, 95%CI:2.01-4.26, P=0.000;HR=2.03, 95%CI:1.37-3.01, P=0.000). Patients with anemia after treatment tended to have a worse PFS and OS (HR=1.59, 95%CI:0.97-2.61, P=0.066;HR=1.60, 95%CI:0.94-2.75, P=0.085). Patients with thrombocytopenia after therapy had a worse OS (HR=2.97, 95%CI:1.01-8.78, P=0.049). Patients with neural toxicity after chemotherapy tended to have a worse PFS (HR=3.36, 95%CI:0.92-12.25, P=0.066). Patients received post treatment after second line therapy had a better OS (HR=0.36, 95%CI:0.22-0.61, P=0.000). Conclusion:Combined with oxaliplatin or cisplatin as second line treatment can’t improve the response and survival in NSCLC patient. Treatment response and PS are prognostic factors to NSCLC patients’ PFS and OS. Patients with treatment related anemia might have a worse survival. Post therapy after failure to second line chemotherapy can prolong the survival.

ABSTRACT

Control of hypertension is essential for stroke prevention. For most patients, the monotherapy often fails to lower blood pressure to normal levels, while the combination therapy can increase the effect of lowering blood pressure, however, not all the antihypertensive drugs have good efficacy. The early data analysis of the Chinese Hypertension Intervention Efficacy Study and the subgroup analysis of the KYOTO HEART study have showed that dihydropyridine calcium channel blocker amlodipine and angiotensin receptor blocker valsartan are both the most effective antihypertensive drugs in the prevention of stroke.

The association of the progress of diabetic nephropathy with the sclerosis of major artery was investigated.Carotid intima-media thickness (CIMT) was positively correlated with urinary albumin and creatinine ratio,and negatively correlated with glomerular filtration rate in diabetes group,suggesting that thickened CIMT may predict the increasing risk of clinical nephropathy in the patients with microalbuminuria.

Objective Comparison on the influence of invigorating spleen and restoring virility to the NPY level in brain and gene expression. Method Establishing the spleen insufficiency model of rat by adopting bitter decline diarrhea, wild eating and overworking, to examine the hypothalamus ventral kernel, the hippocampi CA1 area and forehead cortex, the gene expression change of NPY. Result In the model group, the immunoreaction masculine substance of NPY, hypothalamus ventral kernel,hippocampi CA1 area and prefrontal cortex decreases obviously. The immunoreaction masculine substance of NPY on above mentioned parts in the group of invigorating spleen and restoring virility increases distinctly. In the model group, the expression of NPY mRNA on prefronta cortex and hypothalamus ventral kernel decreases obviously, while on the groups of invigorating spleen and restoring virility increases obviously. Conclusion In the model group, the NPY level and gene expression in brain to promote study memory changed. Invigorating spleen and restoring virility may affect study memory by regulating the NPY level and gene expression.

Gene transfection is an important method for studying gene function. The origination and development of gene transfection are reviewed in this article. Six methods of gene transfection are introduced, among which, special attention are given to two new ones: particle bombardment and photochemistry transfection technology. Besides, the advantages and disadvantages of these methods are compared in order that more suitable methods can be chosen in researches. The application of the technology in gene research is discussed in the end of this article.

ABSTRACT

AIM To study the protective effects and mechanism of chrysophanol on learning and memory impairment of mice with acute senile model induced by AlCl 3. METHODS After sc of AlCl 3 60 mg?kg -1 for 7 d and ip chrysophanol 10,1,0 1 mg?kg -1 for 15 d, using step-through test and step-down test, the effect of chrysophanol on learning and memory was observed and the superoxide dismutase (SOD) activities in cerebrum and glutathione peroxidase (GSH-px)activities in plasma and cerebrum were measured. RESULTS Chrysophanol 10,1,0 1 mg?kg -1 significantly improved me mory impairment induced by AlCl 3 and enhanced the activities of GSH- px and SOD. CONCLUSION Chrysophanol showed protective effect on br ain memory impairment of mice in acute senile model induced by AlCl 3, perhaps the mechanism is involved in enhancing the activities of GSH-px and SOD, cleari ng away the oxygen radicals, protecting the brain neurons from the harm of lipop eroxide.

ABSTRACT

Previous studies have demonstrated that exocytotic release from large dense cored vesicle(LDV)at structurally non-specialized areas within axon terminal of the medullary dorsal horn and it has been speculated that non-synaptic exocytosis from LDV may be a probable mechanism for neuropeptide release. This study provides the evidence that SP containing LDV can release their contents by exocytosis at non-synaptic sites of axon terminal within superficial dorsal horn of medulla oblongata in the rat with ultrastructural immunohistochemistry staining by an unilateral deafferentation. The results of this study confirmed previous hypothesis that SP and perhaps other peptides containing LDV release their contents by exocytosis at non-specialized sites and the significance of such release is discussed.

ABSTRACT

AIM: To elucidate the molecular mechanism of apoptosis in inflammatory cells occurring in the central nervous system during experimental allergic encephalomyelitis (EAE), the changes of pathology and the expression of apoptosis-related molecules Bcl-2 and Bax in the spinal cord of guinea pigs in EAE model were observed. METHODS: EAE model was induced in guinea pig. The cervical and lumbosacral enlargement of spinal cord was obmined during the 14 th and 28 th days after modeling. The H & E staining and ABC immunohistochemistry technique were used. RESULTS: HE staining: The lesion in the model was located in the inflammation of small veins, which were surrounded by inflammatory cells. It was specially noticed that inflammatory cells also infiltrated large veins. The lesions in the 28th day were more severe than that in the 14th day. ABC immunohistochemistry staining: The positive expression of Bcl-2 and Bax was obviously increased in the spinal cord of guinea pig with EAE. The number of Bcl-2 positive cells in the 28th day of model group was more than that of the 14th day. The number of Bax positive cells in the 28th day and 14th day of model group did not show obvious difference at un-lesion areas. CONCLUSION: These results suggest that Bcl-2 and Bax may play an important role in EAE.

The peripheral nerves supplying the acupuncture points of the facial regionwere sectioned from 63 rats and their central terminations were traced by the loss ofthe acid phosphatase(ACP)activity in the substantia gelatinosa(SG)with themodified Gomori's method.1)The supraorbital nerve was cut,the loss of the ACP was confined to thelateral aspect of the substantia gelatinosa in spinal segments C_1.2)The infraorbi-tal nerve was transected,the ACP activity was absent from the central and lateralsectors of the SG in the medulla oblongata.3)The mental nerve was divided,theACP activity was absent from the most medial sector of SG in the medulla oblon-gata and C_1.4)The auriculotemporal nerve was severed,the activity of the enzymewas absent mainly in the inner medial sector of SG in the C_1 and C_2.5)Thefacial nerve was divided,the area deprived of enzyme activity is mainly found inthe central medial sector of SG in C_2.6)The vagal nerve was divided,a medialsector or central sector of SG in the C_1 and C_2 segment is devoid of ACP.7)Thegreat auricular nerve and the cutaneous cervical nerve were severed,the ACP activitywas absent in SG of the upper four cervical cords.The above results show that the primary sensory nerve of the peripheral nerveswhich supply the facial acupuncture points terminate in the SG of the subnucleuscaudalis of the trigeminal nerves and the dorsal horn and the primary sensory nervefibre of these nerves mostly terminate in C_1 and C_2 segment.

ABSTRACT

The segmental and regional projections of the sciatic, tibial and common peroneal nerves to the substantia gelatinosa of spinal cord in rats were studied with the acid phosphatase method. Experiments were carried on 42 rats. Under chloral hydrate anesthesia, sciatic, tibial and common peroneal nerves were severed respecively. Then, after a survival time of 5~12 days, the lumbosacral segments of the spinal cord were removed and sectioned. The frozen sections were incubated with the modified Gomori's method.The projections of the sciatic and tibial nerves to the substantia gelatinosa (SG)were found in L_2~S_1, while these of the common peroneal nerve in L_(2~6).The projections of the sciatic nerve were found in the most medial, the intermediate and the most lateral parts of SG in the segments L_(2~3), and in entire SG in the segments L_4~S_1.The projections of the tibial nerve were shown in the most medial and the intermediate parts of SG in the segments L_(2~5), and in the medial parts of SG at L_6~S_1.The projections of the common peroneal nerve were found in the intermediate parts of SG in all segments L_(2~6).The above study has provided anatomical data for probing into the mechanism of somatosplanchnic interrelations and the principle governing acupuncture anesthesia.

ABSTRACT

The glomerular synapses of the substantia gelatinosa in the spinal trigeminal nucleus of the rat were examined by electron microscopy. The central axonal ending in the glomcruli forms asymmetrical axodendritic synapses on adjacent type 1 and type 2 dendrites. Type 2 dendritic spines or shafts (dendrites which contain synaptic vesicles) form dendrodendritic synapses on Type 1 dendritic spine or shafts (dendrites without synaptic vesicles) and also form dendroaxonic synapses on the central ending. The peripheral terminals (P) form symmetrical axoaxonic synapses on the central ending and form axodendritic synapses on the dendrites in the glomeruli.

ABSTRACT

The substantia gelatinosa of subnucleus caudalis of the trigeminal nerve of the rat shows an intense extralysosomal acid phosphatase activity which disappears after the removal of it's skin of the facial regions.Different regions of the facial skin of 69 rats were excised and the projections of the cutaneous nerves were traced by studying the extralysosomal acid phosphatase activity with the modified Gomori's methods.A somatotopical localization was found within the substantia gelationosa. It agreed with what has been found in earlier studies where other techniques have been used. The findings showed that the fibers of the cutaneous areas along the dorsoventral axis of the body were found in a reverse order in the receptive fields in the substantia gelatinosa. The vertex and fore-head lay in the ventral, while skin of the mandibular areas in the dorsal region, and the skin of the facial areas lay successively from ventral to dorsal in following order: vertex and forehead eye region, dorsal vibrissae, ventral vibrissae-upper lip and finally lower lip-mandibulare region, in the receptive fields of the substantia gelatinosa.The cutaneous facial regions were disproportionately represented in substantia gelatinosa, for example, the field receiving projections from the vibrissae areas or fields were as large as 4mm~2 in the most medial portion of the ventral vibrissae-upperlip which are disproportionately large while the fields receiving projections from the forehead and mandibular cutaneous regions were disproportionately small. The provides a possible explanation for the specific function of analgesia of the acupuncture point Renzhong.

ABSTRACT

The ultrastructural localization of substance P (SP) immunoreactivity, especially the morphology, number and distribution of positive large granular vesicles (LGV) in SP axon terminals of the trigeminal subnucleus caudalis of the rat were studied by electron microscopic immunocytochemistry. This study revealed that SP immunoreactivity was mostly located in axon terminals and unmyelinated fibers. SP axon terminals contained both clear round vesicles and LGV. SP immunoreactivity was found in LGV, and on the surface of clear round vesicles and outer membrane of mitochondria. Positive LGV were spherical or oval in shape (60~120nm in diameter). The number of LGv was mostly 2~3 in a SP axon terminal. LGV often apposed to the axolemma or scattered in the centre of terminal.
